Greg and Kate Brady were heading home to Wauchope along the Oxley Highway at Woolbrook when tragedy struck on Friday afternoon.
Mr Brady was killed almost instantly when his bike collided with a white utility just after 4pm, while Mrs Brady - who was riding pillion - was critically injured when she fell from the motorbike.
A local nurse worked frantically to save the 35-year-old before the Westpac Rescue Helicopter was diverted to the scene after being tasked to Armidale.
Mrs Brady was airlifted to Tamworth Hospital with critical injuries including multiple fractures before later being flown to John Hunter Hospital in Newcastle.
Police confirmed yesterday to The Leader that the 35-year-old had died from her injuries in hospital on Monday morning.
The driver of the utility - and the sole occupant - was not physically injured but was assessed for shock following the ordeal and was taken to Walcha Hospital for mandatory blood and urine tests.
Investigations into the cause of the accident are continuing and a report will be prepared for the coroner.

EARLIER: A MOTORCYCLIST is dead and his female pillion passenger is in a serious condition after a head-on crash near Walcha.
The couple, Greg and Kate Brady, are from Wauchope.
Mr Brady died almost instantly after colliding with the white utility on the Oxley Highway at Woolbrook just after 4pm on Friday.
It’s believed a local nurse rendered aid to Mrs Brady at the scene before the Westpac Rescue Helicopter, which was en route to Armidale, was diverted to the scene.
Ambulance paramedics confirmed Mrs Brady had suffered multiple fractures and head, chest and back injuries, and was airlifted to Tamworth Hospital before being transported to John Hunter Hospital in Newcastle in a serious condition.
Emergency services were on the scene quickly and shut the road, which was littered with debris.
The driver of the ute – the sole occupant – was not injured but was assessed by paramedics as a precaution before being taken to Walcha Hospital for mandatory testing.
The crash closed the road for several hours as investigators tried to work out the circumstances surrounding the crash.
Diversions were put in place around the crash site through Woolbrook but, when dusk fell, the highway was closed at Walcha and at Bendemeer, and vehicles forced to travel via Uralla along the Thunderbolts Way to the New England Highway.
A report will be prepared for the coroner.
