Teams of two or four will navigate through spaces rarely visited and traverse demanding landscapes, moving from checkpoint to checkpoint by foot, mountain bike and kayak.
Race director Richard Old said competitors will have to "problem solve, route find and persevere over a wild course".
"This race will have everything - challenging trek legs, demanding mountain biking - ranging from flowing single track to rugged back country tracks - and scenic paddle legs.
"The course is rich in history and we've designed a race that will give teams a real adventure experience as well as getting to know the area," he said.
The race has attracted top NSW teams BMX Bandits and Over Yonder Racing as well as the owner of the Adventure Racing World Series.
One of the new racers is the all girls team Travel Play Live, from Forster, which is stepping up to 24 hour racing.
Each team will use map and compass, together with their own wits and skills, to navigate their way around an unmarked route by mountain biking, paddling, trekking, orienteering and trail running through the spectacular forests and rivers surrounding Wauchope.
