John William Chardon, 73, was found guilty of manslaughter in September last year after his wife Novy Chardon, 34, disappeared from their Gold Coast mansion in 2013 amid a divorce dispute.
Her body has never been found.
Chardon was sentenced to 15 years in jail.
The Courier-Mail reports Chardon died of a heart attack at Wolston Correctional Centre in Wacol, southwestern Brisbane, on Wednesday night.
Queensland Corrective Services told the publication a 73-year-old prisoner suffered an apparent "medical incident" and paramedics were unable to resuscitate the man.
